Arbidol
Antiviral drug development has led to significant advances in the control of globally prevalent viral infections by the hepatitis C (HCV) and the human immunodeficiency viruses. Nonetheless, there are current gaps in the armamentarium against global viral infections due to some herpes viruses, the hepatitis B virus, and the deadly filovirus Ebola and arenavirus Tacaribe responsible of outbreaks of hemorrhagic fever in emerging countries. Arbidol (ARB) is a cost-effective antiviral with broad-spectrum activity, administered for decades in Russia and China against flu. We showed that ARB inhibits HCV infection, and has potent antiviral activity against the Ebola, Tacaribe, hepatitis B and human herpes-8 viruses. Since these viruses have divergent life cycles, we posit that ARB may exhibit varied modes of action against different viruses. Careful dissection of molecular mechanisms of action of ARB against HCV and the Ebola virus revealed that ARB blocked viral entry. ARB might, therefore, constitute a pharmacological approach against globally prevalent viruses, and an affordable molecule for emerging countries in urgent need for effective antiviral therapies.
Risk of Thrombosis during and after COVID Infection
The novel coronavirus known as SARS-CoV-2 is a respiratory virus that presents with symptoms such as a non-productive cough (60-80%), fever (90%) and shortness of breath (19-40%) and is responsible for the 2019-2020 global coronavirus disease (COVID-19) pandemic. Although most patients experience mild or no symptoms, severe disease is associated with thromboembolic complications and respiratory failure. There is an observed rise in the clinical manifestations of the disease 7 to 14 days after initial symptom onset and also associated with thrombotic complications. Understanding the thrombotic manifestations of the virus is important for patient management.

The integration of micro-fluidics, electrical impedance spectroscopy and stochastic estimation will lead to a device with enhanced detection capabilities. The goal of this thesis was to build a micro-fluidic electrical impedance measurement device that can be used in combination with a stochastic estimator to accurately identify living cells. A microdevice capable of making impedance measurements on individual living cells was designed and built using a series of standard microelectronic fabrication techniques. A microchannel was patterned in SU-8 photoresist between two gold microelectrodes on a two inch Pyrex 7740 wafer. The design process, the fabrication techniques for the microchannel, the fluid port fabrication and the cover slip bonding processes are described in detail. Small glass cover slips were bonded to the wafer using Loctite 3301 adhesive. Impedance measurements of single cells, in the microchannel device, were made using a HP4194A impedance analyzer. Preliminary analysis of the impedance data suggests that Jurkat cells have characteristic impedance signatures, corresponding to their cell type. Cellular Responses of Antioxidants Related to Degenerative Eye Disease Research
Oxidative stress is critical in degenerative retinal diseases. Antioxidants are considered to be potential therapeutics that can treat retinal diseases. Loss of retinal epithelium (RPE) is an important contributor to the degeneration of the retina. Dry AMD induces straight RPE cell degeneration, whereas the wet form causes neovascularization following RPE cell loss. Glaucoma induces progressive retinal ganglion cell (RGC) axon degeneration, resulting in RGC death and visual deficits. RPE cells are also important regulators in eye growth and thus are also susceptible to participating in retinal detachment in myopia. In diabetic retinopathy, the RPE barrier breaks, leading to vascular leakage. The retina is a thin layer in the posterior part of the eye that collects visual information and transfers it to the brain via the optic nerve. Retina consists of RPE cells, photoreceptor rods and cones, glial cells, and neuronal cells, including RGCs. RPE cells support homeostasis by performing a heterocycle of outer photoreceptor segments, enabling nutritional transport from the choroid and removing waste material into the choroid, for example. RPE degeneration leads to vision loss, and antioxidants could protect these cells against oxidative stress, mitochondrial dysfunction, and inflammation, all important contributors to retinal degeneration. This Special Issue is a collection of studies related to cellular responses to oxidative stress and antioxidants in pre-clinical degenerative retinal eye disease research. Oxygen fuels our bodies with life-giving energy - but at a hidden cost: over time, it also slowly degrades the very systems it powers.
