Sarp-Driven Activation of Antibiotic Biosynthetic Gene Clusters in Actinomycetes

Actinomycetes are a group of Gram-positive bacteria of which many representatives are prominent for being prolific producers of bioactive natural products including antibiotics, fungicides, antitumor agents, or immunosuppressants. SARP transcriptional regulators are widely distributed among actinomycetes, especially in streptomycetes and are known to activate antibiotic biosynthesis. The set of genes responsible for the production of natural products, including pathway specific transcriptional regulators such as SARPs, are typically located in contiguous regions of the genome known as "biosynthetic gene clusters" (BGCs). In this book, Oona R繹ssler reports on the activation of antibiotic BGCs in selected actinomycetes strains upon heterologous expression of the SARP-type regulator PapR2 from Streptomyces pristinaespiralis. Applying a bioinformatic screening for the abundance of SARP genes and SARP consensus sequences as part of BGCs, the author has selected actinomycetes candidate strains from the DSMZ strain collection for heterologous SARP expression. It is shown that overexpression of papR2 increased the production of predominantly unknown antimicrobial compounds in more than half of the selected actinomycetes strains, as observed by bioassays against different microbial test strains including bacteria and fungi.

The translation of therapeutics from the lab to the clinic has a dismal record in the fields of neurotrauma and neurological disorders. This is due, in part, to the challenging heterogeneity of the clinical population common to all translational research, but it is also due to the unique challenges of recreating the mechanisms and manifestations of human neurological injury/disorders in small animals. Large animal models are an essential component of successful pipelines for moving discoveries from bench to bedside in other fields (e.g., exploring device or therapeutic scale-up and/or IND/IDE enabling studies), and neuroscience has made significant progress toward establishing such pipelines in its many unique subfields. Due to their size, gyrencephalic brains, high white matter content, and other factors, swine have proven to be ideal for providing high-fidelity, clinically relevant studies to bridge the gap between small animals and humans. Only by recognizing the strengths and limitations of our models can we hope to effectively develop, validate, and translate novel neuroprotective and regenerative therapeutics into clinical practice, in order to maximize functional recovery. Herein, we provide detailed descriptions of and findings from some of the sophisticated swine model systems that have been developed to empower translational research in neurotrauma and neurological disorders.

On the other hand, non-insulated charged conductors generate a dynamic electric field, useful for electrocuting pests. The articles involved explain the structural design of devices and electrostatic principles used for various purposes, including trapping pests and quantitatively analyzing fungal spores, explore the effects of electrostatic fields on pest populations, develop techniques for electrocuting weed seedlings and flies, and establish criteria for target size in trapping and electrocuting pests. These electrostatic approaches complement biological methods, such as breeding pest-resistant crop plants. The reprint includes efforts to screen genetic traits for pest resistance and proposes strategies for controlling viral and fungal diseases. One original article screens resistance traits to Clavibacter michiganensis in wild tomatoes and transfers them to cultivated tomatoes, identifying genetic loci involved in resistance. Biological control measures, including the use of hyperparasitic fungi such Ampelomyces spp., are highlighted for controlling powdery mildew colonies on leaves.

Despite this, much of modern biology teaching tends to be focused on the cellular and molecular level, with the organism often neglected. This accessible textbook provides a general conceptual framework for understanding the organismic level. The book adopts two parallel tracks, with most chapters focusing on one or the other.
