Efficient Screening Techniques to Identify Mutants with Tr 4 Resistance in Banana

Bananas are a staple food for over 500 million people and are also an important cash crop. Fusarium wilt, caused by the fungus Fusarium oxysporum f.sp. cubense, is one of the most destructive diseases of banana globally. Since the 1990s, an aggressive variant of this fungus, called Tropical Race 4 (TR4), severely affected banana plantations in Southeast Asia from where it spread to other continents, including Latin America, where the global banana export market is primarily centred. TR4 is a soil borne pathogen making the disease difficult to contain. The Joint FAO/IAEA Centre of Nuclear Techniques in Food and Agriculture implemented a Coordinated Research Project (CRP) 'Efficient Screening Techniques to Identify Mutants with Disease Resistance for Coffee and Banana" (2015-2020). This CRP brought together experts from Asia, Europe and Africa in addition to experts of the Joint FAO/IAEA Centre to develop resistance against TR4 through mutation-assisted breeding. Induced mutagenesis is particularly attractive in case of banana since most cultivated bananas are seedless, thus hampering conventional cross breeding.This Open Access book is a compilation of the protocols developed under the CRP specifically for TR4. The first part covers methods for mutation induction, including the integrated use of innovative single-cell culture with mutagenesis techniques. The book also describes up-to-date phenotypic screening methods for TR4 resistance in banana under field-, greenhouse- and laboratory conditions. Finally, molecular and bioinformatics tools for genome-wide mutation discovery following Next Generation Sequencing are also described. Given the imminent threat of Fusarium Wilt TR4 on banana production globally, it is our hope and intention that the book will serve as a timely reference and guide for banana breeders and pathologists worldwide who are committed to the genetic improvement of banana for Fusarium wilt resistance.

The genus Diospyros contains about 400 species distributed all continents of the globe. Most of Diospyros species are distributed in the tropical and subtropical regions, while cultivated persimmon or simply kaki (Diospyros kaki) originates in East Asia and has been grown in China, Japan and Korea for many years. World production of persimmon has been increasing continuously since 1990s, with China being the most producer. Outside Asia, kaki production has been rapidly increasing in Spain in recent years. Because kaki is mostly hexaploid (2n = 6x =90) and its genome size is quite large with the flow cytometric analysis estimation of about 900 Mb, genetic and molecular studies in persimmon are quite complicated. Diploid close relatives of kaki, D. lotus and D. oleifera have been recently used as model species of kaki. Utilizing these species, molecular basis of several economically molecular basis of important traits such as sexuality and astringency has been studied, giving important information for kaki breeding.

Of the global population of more than 7 billion people, some 800 million do not have enough to eat today. By 2050, the population is expected to exceed 9 billion. It has been estimated that some 15% of food production is lost to plant diseases; in developing countries losses may be much higher. Historically, plant diseases have had catastrophic impact on food production. For example: potato blight caused the Irish famine in 1845; brown spot of rice caused the Great Bengal Famine of 1943; southern corn leaf blight caused a devastating epidemic on the US corn crop in 1970. A Flora of the Trinity Alps of Northern California offers a thorough botanical study of one of California's most unique and rugged regions. Ozark Plants is a full-color field guide to over 500 plant species found in the Ozark Mountain region of southern Missouri, northern Arkansas, and the northeastern corner of Oklahoma. Illustrated with hundreds of color photographs, the book describes each plant followed by a discussion of the plant's distribution, habitat, and value for wildlife and livestock. A key is provided so that the reader may more easily identify unknown plant specimens. The Ozark region is a plateau and mountain area of more than 40,000 square miles at the western edge of the eastern deciduous forest, and host a rich assemblage of plant species. Overall, nearly 3,300 vascular plant species are known from the region, within approximately 1066 genera and 186 families. This diversity is due, in part, to a long presence uninterrupted by glaciation, varied site and soil conditions, and a midcontinental geographic position. This geographical location has resulted in many species with distributional centers outside of the Ozarks reaching their range limits here. For example, the range of certain plants from the southwestern United States extends to the Ozarks, typically in dry-soil environments such as the glades. On moist, north-facing slopes, plants from the northern and eastern United States may be found. Some typically southern species occur in special habitats, including many parts of the Boston Mountains.
