Resilient Forest Management

Global forest management is now grappling with ways to address the many dimensions of global change, including a warming climate and increasing forest disturbance from fires and pest outbreaks, along with changes in public values. However, the dominant forest management paradigms still assume a constant and predictable world in which command-and-control (i.e., treating long-lived forests much like short-lived agricultural crops) and single-value (i.e., timber) optimization still prevail. This novel text argues for new approaches to forest management that focus on resilience and embracing adaptability to the changing socio-ecological environment as it unfolds. Resilience is the ability of a system to maintain its essential attributes (in the form of composition, structure, and/or function) in response to stress, disruption, or disturbance. Managing a system for resilience places an emphasis on persistence rather than growth, efficiency, or profitability, which can be fulfilled by enhancing the capacity to resist change (i.e., robustness) or by enhancing the capacity to incorporate change in desirable directions (i.e., flexibility), or a combination of the two. Resilient Forest Management develops many of the same resilience-enhancing strategies for protected areas, multi-purpose forests, and timber production lands, but with different degrees of emphasis. Featured prominently are practices that enhance diversity, connectivity in space and time, and adaptive management as informed by vulnerability analysis and broad stakeholder consultation. In so doing, Resilient Forest Management builds on foundational concepts of ecological forestry and our understanding of complex adaptive systems and takes sustainable forest management to the next level. Ewing Sarcoma

Ewing sarcomas are characterized by oncogenic ews/ets translocations. However, these oncofusions do not determine the outcome. Patient fate is determined by metastasis, and the complex spreading process is far from being completely understood. Ewing sarcomas are childhood cancers with a low mutational rate, making biomarkers elusive. Mutations increase with relapse and mutagenic therapy exposure. A silent tumor genome limits targeted therapy approaches. Nevertheless, precision oncology approaches aim to enhance the therapeutic index of conventional chemotherapy with novel small molecules targeting epigenetics, metabolism, and stress responses. Although their genome is generally silent, Ewing sarcomas reactivate endogenous retro-elements. Their activation is linked to an inflammatory response and a prometastatic modulation of the microenvironment. Moreover, in most cases, these tumors are characterized by a scarcity of T cell infiltrates and a predominance of immunosuppressive myeloid cells, leading to inflammation and immunosuppression. These suppressive myeloid cells shield the tumor against adaptive immune attack, hampering the efficacy of chimeric antigen receptor or T cell receptor transgenic T cells. Overcoming these immunosuppressive mechanisms may enhance immunotherapeutic efficacy. One approach is the utilization of oncolytic viruses, which are genetically engineered to depend on their lytic cycle from metastatic drivers. They can induce immunogenic cell death and, particularly when in combination with cell cycle inhibitors, also have the potential to overcome barriers to immunotherapy.

Zebrafish

Zebrafish are an attractive vertebrate model used in drug discovery. Due to their small size, external development, and transparency, zebrafish embryos are compatible with multiwell plates for high-throughput screening. Based on the availability of zebrafish genome sequences, the ease of creating transgenic and genetic mutants, and the conservation of signal transduction pathways, a variety of human diseases have been modeled using zebrafish. Moreover, biological pathways, such as physiological and molecular events, which control the development and function of most organ systems, including the cardiovascular, skeletal, nervous, digestive, and visual systems of zebrafish, are similar to those in mammals. The zebrafish reference genome published in 2013 has further accelerated the use of zebrafish in human disease modeling. Over the last two decades, zebrafish have become popular in pharmaceutical and toxicological research. Consequently, several drugs used to treat human diseases (including tuberculosis, neuronal, and auditory disorders, as well as several types of cancer) have been identified from zebrafish screens. Such advances in this area of research have established zebrafish as an invaluable human disease model. This collection of articles aims to cover current research and provide an overview of the advances made in the area of drug discovery and disease modeling using the zebrafish model.

The prevalence of obesity continues to rise worldwide, contributing to an increased burden of metabolic disorders such as type 2 diabetes, dyslipidemia, and cardiovascular diseases.
