State-of-Art in Innate Immunity
The innate immune system is the first line of defense against bacterial and viral infections and sterile inflammation through the recognition of pathogen-associated molecular patterns (PAMPs) by pathogen recognition receptors (PRRs) resulting in the production of proinflammatory and antiviral cytokines and chemokines. Several damage-associated molecular patterns (DAMPs), which were released by passive or active mechanisms under sterile conditions, are additionally recognized by PRRs and can cause or even aggravate the inflammatory response.In this special issue many aspects of innate immunity are summarized. Mechanisms of different DAMPs to induce pro- and anti-inflammatory activities, functions of different immune cells, as well as the crosstalk between coagulation and innate immunity were described. Furthermore, aspects of autoinflammatory diseases, types of programmed cell death pathways, and insect immunity are covered. Finally, therapeutic options for the treatment of diseases related to autoimmunity or infections are suggested. Overall, this special issue presents a broad overview of activities related to sterile inflammation and defense mechanisms of innate immunity.

An Ex Vivo 3D Tumor Microenvironment-Mimicry Culture to Study TAM Modulation
Tumor-associated macrophages (TAMs) accumulate in the solid tumor microenvironment (TME) and have been shown to promote tumor growth and dampen antitumor immune responses. TAM-mediated suppression of T-cell antitumor reactivity is considered to be a major obstacle for many immunotherapies, including immune checkpoint blockade and adoptive T/CAR-T-cell therapies. An ex vivo culture system closely mimicking the TME can greatly facilitate the study of cancer immunotherapies. Here, we report the development of a 3D TME-mimicry culture that is comprised of the three major components of a human TME, including human tumor cells, TAMs, and tumor antigen-specific T cells. This TME-mimicry culture can readout the TAM-mediated suppression of T-cell antitumor reactivity, and therefore can be used to study TAM modulation of T-cell-based cancer immunotherapy. As a proof-of-principle, the studies of a PD-1/PD-L1 blockade therapy and a MAO-A blockade therapy were performed and validated.

Neural Control Engineering
How powerful new methods in nonlinear control engineering can be applied to neuroscience, from fundamental model formulation to advanced medical applications. Ethics in Everyday Places
An exploration of moral stress, distress, and injuries inherent in modern society through the maps that pervade academic and public communications worlds. Effective Public Health Policy in Organ Donation
This book shows how a successful public health policy designed to foster the culture of brain death diagnosis and improve the interaction between Organ Procurement Organizations and Intra-Hospital Organ and Tissue Donation Committees for Transplants led the Brazilian state of Paran獺 to reach one of the highest rates of effective organ donors per million population in the world. Brazil has the largest public transplant system in the world, and each of the country's 26 states is responsible for organizing its own transplant system. Madness in the Family
To fend off American and European imperialism in the nineteenth century, Japan strove to strengthen itself by drawing on the most updated ideas and practices from around the world. By the 1880s, this included the introduction of Western-derived psychiatry and its ideas about mental illness. The first Japanese psychiatrists claimed that mental illnesses required medical treatment in specialized institutions rather than confinement at home, as had been common practice. Yet the state implemented no social welfare policies to make new medical services more accessible and affordable to the public. The family, especially women, thus continued to carry the burden of caring for those considered mad. Madness in the Family examines how the family in Japan came to be seen as the natural provider of care for those suffering from mental illnesses. It centers on the experiences of women and families, which have long been obscured by the voices of male psychiatrists, state officials, and lawmakers. H. Yumi Kim traces how women and families negotiated a dizzying array of claims about madness and its proper management across various settings. In the countryside, psychiatrists tried to refute the notion that fox spirits could cause madness, and the government regulated the use of cage-like structures inside homes. In cities, a booming medical marketplace spread ideas about feminized illnesses such as hysteria, and female defendants were evaluated for menstruation-induced disorders. As women and families navigated this shifting therapeutic landscape, they produced their own gendered approaches to madness that would take precedence over the claims of psychiatry, the law, and the state in everyday life. Decoupling the history of mental illness from the discipline and institutions of psychiatry, Madness in the Family reveals the power and fragilities of gender, kinship, and care in the creation of different modes of caring for and understanding mental illness that persist to this day.

Quantitative Decisions in Drug Development
This book focuses on important decision points and evidence needed for making decisions at these points during the development of a new drug. It takes a holistic approach towards drug development by incorporating explicitly knowledge learned from the earlier part of the development and available historical information into decisions at later stages. In addition, the book shares lessons learned from several select examples published in the literature since the publication of the first edition. The second edition reiterates the need for making evidence-based Go/No Go decisions in drug development discussed in the first edition. It substantially expands several topics that have seen great advances since the publication of the first edition. The most noticeable additions include three adaptive trials conducted in recent years that offer excellent learning opportunities, the use of historical data in the design and analysis of clinical trials, and extending decision criteria tothe cases when the primary endpoint is binary. The examples used to illustrate the additional materials all come from real trials with some post-trial reflections offered by the authors. The book begins with an overview of product development and regulatory approval pathways. It then discusses how to incorporate prior knowledge into study design and decision making at different stages of drug development. Prior knowledge includes information pertaining to historical controls. To assist decision making, the book discusses appropriate metrics and the formulation of go/no-go decisions for progressing a drug candidate to the next development stage. Using the concept of the positive predictive value in the field of diagnostics, the book leads readers to the assessment of the probability that an investigational product is effective given positive study outcomes. Lastly, the book points out common mistakes made by drug developers under the current drug-development paradigm. The book offers useful insights to statisticians, clinicians, regulatory affairs managers and decision-makers in the pharmaceutical industry who have a basic understanding of the drug-development process and the clinical trials conducted to support drug-marketing authorization. The authors provide software codes for select analytical approaches discussed in the book.
