Recent Advances in Psychopathology in Neurodevelopmental Disorders
Psychopathology, including functional psychiatric disorders, such as schizophrenia, bipolar disorder, major depressive disorder and anxiety disorders, and also dementia and challenging behaviour, is common in people with neurodevelopmental disorders (NDDs), such as intellectual and developmental disabilities (IDD), autism spectrum disorder (ASD) and attention deficit hyperactivity disorder (ADHD), and is the cause for a major public health concern. Much research has been conducted in the last three decades to understand this issue and develop effective interventions. Still, a lot remains unknown, for which high-quality translational research is urgently needed. In this Special Issue, we drew together research on recent advances addressing these issues. The areas covered included life expectancy among people with and without intellectual disabilities, the social and emotional development of people with intellectual disabilities and ASD, the influence of external environmental factors in determining aggressive behaviour in people with intellectual disabilities, parent and child training for children with autism and intellectual disabilities, how the presence of deafness influences quality of life, the views of prescribers on the deprescribing of antipsychotics among adults with intellectual disabilities, the views of people with intellectual disabilities who took part in the deprescribing, the views of direct-care workers on the causes of challenging behaviours in people with intellectual disabilities, and the validation of tools to detect cognitive decline in adults with intellectual disabilities.
Targeting Inflammation and Inflammatory-Related Diseases with Natural Bioactives
Inflammation is a complex biological response to injury as a result of different stimuli such as pathogens, damaged cells, or irritants. Inflammatory injuries induce the release of a variety of systemic mediators, cytokines, and chemokines that orchestrate cellular infiltration, consequentially bringing about the resolution of inflammatory responses and the restoration of tissue integrity. However, persistent inflammatory stimuli or the disregulation of the mechanisms of the resolution phase can lead to chronic inflammation and inflammatory-based diseases. Nowadays, commercially approved anti-inflammatory drugs are represented by nonsteroidal anti-inflammatory drugs (NSAID); glucocorticoids (SAID); and, in some cases, immunosuppressant and/or biological drugs. These agents are effective for the relief of the main inflammatory symptoms. However, they induce severe side effects, and most of them are inadequate for chronic use. Starting from these premises, the demand for new, effective, and safe anti-inflammatory drugs has led research in new therapeutic directions. The recent and emerging scientific community slant is oriented towards natural products/compounds that could represent a boon for the discovery of new active molecules and for the development of new drugs and potentially useful therapeutic agents in different inflammatory-related diseases. We hope that it will stimulate the interest of the scientific community involved in studying the effects of natural and synthetic compounds in different fields of interest such as acute and chronic inflammation, inflammatory pain, inflammation-related diseases (e.g., autoimmune diseases), amongst others.

Model-Informed Precision Dosing
Model-informed precision dosing (MIPD) is an advanced quantitative approach focusing on individualized treatment optimization. MIPD integrates mathematical models of drugs and diseases combined with individual patient characteristics (e.g., genotype, anthropometric factors, and organ function). MIPD has been highlighted as a useful tool for drug dosage selection in both the drug development process and clinical practice and it is a rapidly growing discipline that is supported by the main drug regulatory agencies. Despite the potential benefits of this methodology toward personalized medicine, its application is still limited. The Special Issue presented here includes several PKPD and PBPK models focused on improving the current state of art regarding the PK behaviour of different drugs with the aim of improving the efficacy/safety balance of these treatments and their clinical outcome; the Special Issue is intended to be of particular interest for clinical pharmacologists, pharmacometricians, and specific clinicians who routinely use the considered drugs.

Mental Health Challenges during the COVID-19 Pandemic
The aim of the reprint was to provide readers with updated research findings on the psychological and psychiatric consequences of the pandemic. Optic Neuropathies
Optic neuropathies are conditions in which there is damage to the optic nerve (ON) caused by a variety of causes, including glaucoma, inflammation, gene abnormalities, ischemia, trauma, and toxicity. ON damage triggers a process of axon degeneration, inflammatory cytokine upregulation, breakdown of the blood-optic nerve barrier, and, eventually, the induction of apoptosis of retinal ganglion cells (RGCs), resulting in optic atrophy. To date, there is no effective treatment for most optic neuropathies; however, because the damage initially is axogenic, there may exist a window of therapeutic opportunity before the death of RGCs. Thus, the search for effective treatments for various optic neuropathies before there is permanent damage to prevent or limit visual dysfunction and the development of methods to stimulate axon and/or RGC regeneration to restore vision after damage has occurred is pivotal.This reprint collected 19 articles published in this Special Issue of IJMS which covers the molecular mechanisms to protect RGCs and/or axonal damage, translational research, gene therapy, regenerative medicine, and neuroprotection for glaucoma.

Challenges and Opportunities for Effective Cancer Immunotherapies
Anti-cancer immunotherapies have generated spectacular outcomes in the clinical environment and changed treatment schemes for cancer patients. Adoptive cell therapies (ACTs), including using autologous tumor-infiltrating lymphocytes and chimeric antigen receptor (CAR) T cells, and checkpoint blockades have emerged as the most effective treatments for certain cancers. The current challenge for cancer immunotherapies is that although some patients have benefited from the treatments, a number of cancers are resistant. The purpose of this Special Issue is to understand anti-cancer immunotherapy treatment resistance mechanisms and explore new options to provide opportunities for effective treatments.

Recent Advances in Child and Adolescent Psychiatry
Psychiatric disorders in children and adolescents are relevant both in terms of their prevalence (they are roughly estimated to affect about 20% of school-aged children and adolescents) and of their impact on the quality of life of the child/adolescent and their family, but also on the society as a whole. Disorders affecting children and adolescents are increasingly seen as having highly specific features, and, therefore, require an equally specific approach despite the coexisting need to integrate contributions from other professions (e.g., pediatricians, psychiatrists, psychologists, and therapists of different sorts). However, the available data are still insufficient, especially for treatment options and too often therapeutic choices are still made based on studies conducted in adults. Even more important, the existence itself of some disorders is questioned especially outside the scientific field (e.g., ADHD). Recent Progress of Nanomedicine and Targeted Drug Delivery for Cancer Treatment
Currently, cancer is the second leading cause of death worldwide and is the most complex and challenging disease known to humankind. Due to the complex underlying mechanisms of tumorigenesis and tumor metastasis, as well as the intrinsic limitations of conventional cancer therapies, the effective treatment of cancer remains a great challenge. Novel therapeutic strategies with superior anticancer ability urgently need to be discovered. In recent years, nano-drug delivery systems have been extensively investigated and applied as alternatives to conventional cancer treatments. Cancer nanomedicines aim to achieve the targeted delivery of chemotherapeutic drugs to the tumor site, utilizing strategies such as passive targeting, active targeting and stimuli-triggered drug release, while simultaneously decreasing drug accumulation in normal tissues, which together lead to improved therapeutic efficacy and reduced side effects. Currently, several cancer nanomedicines are in regular use, and several others are in various stages of development.This Special Issue will highlight recent progress in the development of novel nanomedicines and targeted drug delivery systems for cancer therapy.
Novel Therapeutic Considerations in Bone and Soft Tissue Sarcoma
Sarcomas are a group of rare cancers that arise in connective tissues of the body. Surgical resection is pivotal for the management of locoregional disease. In locally advanced or metastatic disease settings, systemic therapy has an important role in the multidisciplinary management of sarcoma. Cytotoxic therapy has been the primary treatment for many years. However, recent advances in molecular pathogenesis, the investigation of the tumour microenvironment, changes in clinical trial design, and increased international collaboration have led to the development of histology-driven therapy. Furthermore, genomic profiling has highlighted that, while some sarcomas have complex karyotypes, others are driven by translocation, amplification, and mutation, representing targets for the development of novel therapies. Checkpoint inhibitors have been used as single agents or in combination in clinical sarcoma trials. This progress will move the therapeutic modality in sarcoma patients from the "one-size-fits-all" approach towards a more personalized therapeutic algorithm and better outcomes soon. In this Special Issue, we present original research and review articles highlighting novel therapeutic approaches in the treatment of sarcoma patients.
Gender Difference in Diabetes
Sex and gender can affect the incidence, prevalence, symptoms, course and response to drug therapy in many illnesses, with sex (the biological side) and gender (the social‒cultural one) being variously interconnected. During the two days of the meeting "Gender Differences in Diabetic Disease" held in Olbia, Italy, on 4 and 5 December 2020, we discussed the various medical, social, ethnic, psychological and anthropological aspects of gender differences to build as broad a picture as possible of how these differences are present and evident in diabetic disease. Type 2 diabetes is the perfect example to justify gender medicine. During the meeting, we also tried to identify the still-unclear points that deserve further studies.This book describes several aspects of gender medicine that are related to diabetes. Physiopathology, drug treatment, sexual and psychological aspects, diabetes complication, migration, burden of working with a chronic disease and aspects related to COVID-19 infection are explored in a gender differentiation way. Human Papillomavirus and Head and Neck Cancer
In the study of head and neck squamous cell carcinoma (HNSCC), we know that virally induced malignancies behave differently than those generated through other carcinogenic mechanisms. This paradigm extends into differences in tumor growth, treatment response and resistance, as well as invasion and metastasis. The inability to correctly diagnose and appropriately treat HNSCC at early stages has contributed to reduced survival for HNSCC patients, particularly those with HPV-negative disease. The examination of the molecular differences between HPV-positive and HPV-negative HNSCC will provide support for early diagnostic markers and guide appropriate treatment selection to improve patient care and survival.This Special Issue highlights current research efforts to understand the evolving contribution of human papillomavirus in HNSCC.
